I have a usb based- usbtiny and avrisp mkI programmer(both have a 10 way idc cable compatible with the 10 pin header on the mjoy16).
To program the mjoy16 I need to connect the idc cable to the mjoy isp and the other end of the programmer to the usb.
Do I need to plug the mjoy16 to the usb separately?

Also do I need the 330ohms resistors(which are on the original mjoy16 pcb)?

Is programing the mcu separately on a target board a better option?

You should not need 330ohm resistors with usbtiny or avris mk1.

Doesn't matter where you program it. If your programmers supply 5V, then you don't need USB.
